What is Semax? A plain-English guide
Where the peptide came from, what it does in the brain, and what thirty years of research actually measured.
01The short version
Semax is a short synthetic peptide, seven amino acids long, that has been studied for attention, memory and learning since the early 1990s. It is usually taken as a nasal spray, it contains no caffeine or other stimulant, and it is the only active ingredient in FOCUS-01.
It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, registered for clinical use in Russia in 1994, and has been in continuous use there since. Outside Russia it sits in an odd position: well studied for its class, widely used, but not licensed as a medicine in the UK, the EU or the US.

02What Semax actually is
The sequence is Met-Glu-His-Phe-Pro-Gly-Pro. The first four amino acids are a fragment of adrenocorticotropic hormone (ACTH), specifically positions 4 to 7. Researchers in the 1980s noticed that short fragments of ACTH influenced attention and learning in animals without producing any of the hormone’s effects on the adrenal glands. The fragment carried the cognitive activity but not the hormonal baggage.
The problem with a bare fragment is that the body breaks it down in minutes. The solution was to attach a three-amino-acid tail, Pro-Gly-Pro, which slows that breakdown and lets the molecule stay active for hours rather than minutes. ACTH(4–7) plus PGP is Semax.
So it is not a plant extract, not a vitamin, and not a stimulant. It is a designed molecule with one job, which is why it can be sold as a single compound at a stated purity rather than hidden inside a blend.
03What it does in the brain
Three findings come up again and again across the literature.
1. It raises BDNF
Brain-derived neurotrophic factor is the growth factor the brain uses to strengthen connections between neurons. It is central to learning and memory consolidation. In rat brain tissue, a single dose of Semax raised BDNF protein 1.4-fold in the hippocampus and increased the activated form of its receptor, TrkB, 1.6-fold [3]. In glial cell cultures the effect on BDNF messenger RNA was roughly eight-fold within hours [2]. Follow-up work found the same pattern in the hippocampus, frontal cortex and basal forebrain [5][6]. This is the most replicated finding about Semax and the most likely explanation for its effects on memory and learning.
2. It modulates dopamine and serotonin
Rather than flooding the brain with a stimulant, Semax was shown in rodents to activate the brain’s own dopamine and serotonin systems [4]. This is a plausible route to drive and motivation that does not involve raising heart rate or blood pressure, which is why users describe the effect as quiet rather than jittery.
3. It changed attention and memory scores in people
The human evidence rests mainly on one controlled study from 1996. Healthy volunteers were tested across eight-hour working shifts with and without an intranasal dose of Semax (0.25 to 1 mg). Attention and short-term memory scores improved, the effect was largest in the people who were most tired, and improved work efficiency was recorded for 20 to 24 hours after a single dose [1].

04Why it is a nasal spray
Peptides do not survive the digestive system. Stomach acid and digestive enzymes break them apart, and whatever survives that meets the liver before reaching general circulation. Swallowing Semax would waste most of it.
The nasal cavity is thin-walled and densely supplied with blood vessels, so a compound delivered there crosses into circulation directly. Every human and animal study of Semax used the intranasal route, which is why FOCUS-01 is a metered spray and not a capsule. A pump actuator also delivers the same volume every press, so one spray per nostril is one dose without any measuring.
05How people use it
The pattern that fits the research is a single dose 20 to 30 minutes before the work you want to be sharp for: before the market opens, before a deep-work block, before an exam. Most people use it once a day. FOCUS-01’s directions are one spray per nostril, and never more than four sprays in 24 hours.
Because there is no stimulant, there is no peak-and-crash curve, and it does not double up with coffee. Timing, duration and storage are covered properly in the dosage guide.
06Where the evidence stops
Be clear-eyed about this. The human study is small and from 1996. The mechanistic work is in rats and cell cultures. Much of the literature was produced in Russia under different research standards, and there are no large, modern, independent trials in healthy adults. Long-term safety data in healthy people is thin. Semax is not a licensed medicine in the UK and is not a treatment for any condition.
That is the honest state of the evidence. It is stronger than almost anything sold as a nootropic, and weaker than a prescription drug that has been through phase III trials. We say so on every page because a customer who knows what they are buying is a customer who comes back.
