BDNF: what it is, why it matters for learning, and how to raise it

The brain's growth signal, what raises it (exercise, sleep, Semax) and why there is no BDNF supplement as such.

01The short version

BDNF, brain-derived neurotrophic factor, is the protein the brain uses to grow and strengthen the connections between neurons. More of it in the right places means faster learning and better memory consolidation; less of it is linked with low mood and cognitive decline. Exercise, sleep and a few compounds raise it. Semax is one of the few that does so quickly, which is the mechanism behind its effect on memory.

02What BDNF does

Every time you learn something, the synapses involved are physically changed: new receptors, new spines on the dendrites, sometimes new connections. BDNF is the signal that authorises and fuels that construction. It binds to a receptor called TrkB on the neuron and sets off the cascade that makes the change stick. Without enough of it, short-term memories fail to become long-term ones. Researchers sometimes call it "fertiliser for the brain", which is a fair picture.

03What raises BDNF

Exercise

The best-evidenced method. Aerobic exercise raises circulating BDNF within a session and baseline levels over weeks. This is the mechanism behind "a run clears my head".

Sleep

Sleep deprivation lowers BDNF; recovering sleep restores it. Another reason no supplement fixes a short night.

Diet and fasting

Omega-3 intake and intermittent fasting both raise BDNF in animal studies, with weaker human data.

Semax

In rat hippocampus, a single intranasal dose raised BDNF protein 1.4-fold and TrkB 1.6-fold [3]. In glial cultures BDNF mRNA rose about eight-fold within hours [2], and the same rise was found in the frontal cortex and basal forebrain [5][6]. It is the most replicated finding about the molecule and the likely basis of the memory effects seen in people [1].

Others with some evidence

Curcumin, Lion's Mane and Noopept all have animal data; none has convincing human evidence for raising BDNF.

04Is there a BDNF supplement?

Not directly. BDNF is a protein; swallowed, it is digested, and it does not cross the blood-brain barrier from the blood anyway. What exists are things that make the brain produce more of its own: the list above. Products sold as "BDNF supplements" are usually a mushroom extract or a blend, and they are relying on animal data. If you want the fastest and best-documented pharmacological route, it is Semax by the nasal route, alongside the exercise and sleep that do most of the work.

05The honest caveat

Almost all BDNF measurement is in animals or in blood, because measuring it in a living human brain is not possible. The link between "raises BDNF in rats" and "you remember more" is strong in theory and consistent with Semax's human study, but it is inference, and we present it as such on our research page.
