Peptide nasal sprays explained: why peptides go up the nose

01The short version

Peptides are chains of amino acids, and the body treats a swallowed one as food: stomach acid and digestive enzymes take it apart before it reaches the blood. That is why insulin is injected and why the nootropic peptides Semax and Selank are taken as nasal sprays. The nose is the one route that is quick, painless, needle-free and gets the molecule to the brain intact.

02Why not a capsule

Three barriers stand between a swallowed peptide and your brain. The stomach's acid unfolds it. Enzymes in the gut cut it into fragments. Whatever survives passes through the liver before general circulation, and the liver removes most of it. For a seven-amino-acid peptide like Semax the fraction that reaches the brain by mouth is close to nothing. Every study of Semax, in animals and in people, used the intranasal route for this reason [1].

03Why the nose works

The nasal cavity has two things a peptide needs. A large, thin, blood-rich lining that absorbs small molecules directly into circulation, skipping the gut and the liver. And, at the top, the olfactory region, where nerve endings run straight from the nasal lining into the brain. Molecules delivered there can travel along those nerves and around them, which is a route past the blood-brain barrier that no capsule has.

The result is onset in minutes rather than the hour a tablet takes, a usable fraction of the dose, and no needle. Technique matters, which is why we wrote how to use a nasal spray properly.

04Which peptides are used this way

• Oxytocin: the best-known intranasal peptide, used in research on social behaviour.
• Desmopressin: a licensed nasal peptide medicine, which is a useful proof that the route is established pharmacology rather than a hack.
• Insulin: intranasal insulin is under study for memory in older adults.

Peptides such as BPC-157 and the GLP-1 drugs, which dominate the "peptides UK" conversation, are injected, not sprayed; the molecules and the goals are different, and they are not what COGENT makes.

05What a good peptide spray needs

A stated purity for the peptide, assayed per batch. A sterile, preserved solution so it does not grow anything in the month it takes to use. A metered pump so each press is the same dose.
